A prodrug is a "compound that undergoes transformation before exhibiting pharmacological effects" (IUPAC). In other words, it is a reformulation of an existing drug that changes how and where the drug takes effect in the body. Valaciclovir, for example, is absorbed by cells and converted to aciclovir within the cell, making it more orally bioavailable; aciclovir, in turn, is now limited largely to topical use.
